以前啊如果要扩增DNA,就得繁殖很多很多的那小小的细胞(细胞带有目的DNA,属于体内扩增)。
后来有个叫Kary Mullis博士的家伙跑出来说,你其实可以在体外很简单的达到目的。
你只要把你的模板DNA(就是带有目的DNA的DNA)跟缓冲液混匀了,当然也要加一些引物,核苷和聚合酶。
然后,变性(94度,让DNA双链变性成单链),退火(让引物可以跟匹配的DNA序列结合,温度在五六十度左右),延伸(72度,聚合酶的工作温度,目的DNA片断被复制),你会发现加加热降降温也会有奇妙的效果。
当你想检测突变时,来做PCR吧;
当你想重组基因时,来做PCR吧;
当你要亲子鉴定时,来做PCR吧;
当你要解决案件时,来做PCR吧。
